The anthemic performance included a few costume changes, a guest appearance from Tame Impala’s Kevin Parker, and flashy dance moves.
However, fans watching the BBC’s iPlayer found that the audio was choppy on many issues, slow on others, and often flat.
Angry fans took to social media to criticize the 28-year-old artist for allowing her to be unwell during one of the greatest performances of her era.
One said: “The sound on the BBC stream of #Glastonbury is… weird. This Dua set sounds flat.”
Others posted: “Dua Lipa – either she’s copying or her sound mix is so wrong. It just sounds bad.”
Future A Third wrote: “Can someone please turn Dua Lipa’s mic up? Can barely hear her! The sound is terrible every year!”
Dua produced hits with Kevin Parker including Be the One, Hallucinate, Unutilized Regulation and Tame Impala’s The Less I Know the Higher.
Introducing Parker, Dua said: “This is a guy I admire and who I asked to work with on my album Radical Optimism. Tonight I get to pretend to be in Tame Impala … Please welcome Kevin Parker.”
Dua said that she had dreamed of starring in the Glastonbury competition “her whole life”.
She said: “I can’t believe it, I’ve dreamed about this my whole life. And I can’t believe I’m right here, right here, with you guys, I’m so happy to be here with you guys tonight.”
Others accused Dua of cheating during the set that started with the training season.
One person said: “”What a shame #Glastonbury has a headline act who is clearly copying her singing.”
But fans came to her defense, saying: “No, Dua Lipa is not copying. Most of the lead vocal parts are absolutely clearly live. She’s tearing it up and everyone on stage looks like they’re having the time of their lives.” “Enjoying it.”
The Brit-Albanians released their third album Radical Optimism earlier this year and won Best Pop Act at the Brits.
In the lead-up to his set he revealed what the secret to perfecting his choreo-heavy performance was.
He told the podcast Dish From Waitrose: “I haven’t done any proper tour shows for a year, so I need to get my stamina back before I go to festivals.”
The singer explained, it involved “running, dancing, singing all at the same time, to make sure I could hack it once the adrenaline hit.”
“It’s like rehearsal, no drinking, no working out, just constantly preparing for all the upcoming shows.”
Saturday night at Glastonbury is set to see Coldplay headline the Pyramid Stage with Little Simz opposite them.
Other acts scheduled to perform today include Camila Cabello, Peggy Gou and Disclosure.
SZA will close the festival tomorrow, with the Americans performing followed by Shania Twain and Burna Boy.
Hugh British band Kasabian will also return to Worthy Farm this weekend with a hit-packed set led by frontman Serge Pizzorno.
Insiders confirmed that they have been booked to play the Woodsies on Saturday at 6pm.
A source said: “Glastonbury is always a big deal but this year Kasabian really can’t wait, especially since it’s been a decade since they were last there.”

Dua Lipa was rehearsing for her Glastonbury set at a venue in Birmingham when a TikToker tried to break in.
The levitating singer was working on her routine with her team of dancers when the man arrived on site and then tried to stroll into the rehearsal space during filming.
A source said: “Dua has been working very hard over the last few days to get her set right for Glastonbury. When his team reached the site on Friday, he sent the man away.
“He ruined his entire image by trying to make a film and it was all quite shameful.
“Some people from Dua’s team spoke to him and asked him to clear the matter. She was inside so luckily all the noise was missed. Nothing is going to throw him off course this weekend. Dua can’t wait to show Glastonbury what she’s been working on.
Superstar’s Glasto promises to be a spectacular weekend of live tracks from headliners Coldplay, SZA and legends Shania Twain.
Dua’s eagerness to make plans has been going on for a long time.
We first learned in October how she was being battled by Glasto boss Emily Avis and US Coachella heads.
Emily shared with Dua related resources how she wanted to approach the at-home scenario for her trusted lovers.
